Association of the severity of tongue lesions in broiler chickens with the quality of corn in feed PAB
Vaccari,Isabel Cristina Mores; Duarte,Vinicius; Rodrigues,Denise Russi; Santos,Fabiana Ramos dos; Minafra,Cibele Silva.
Abstract: The objective of this work was to determine the correlation of the physical attributes of corn and the mycotoxins present in its grains with the severity of tongue lesions in broiler breeder roosters. The occurrence of the aflatoxin, vomitoxin, zearalenone, T-2 toxin, and fumonisin mycotoxins was detected in the corn grains used in the feed formulation for breeder chickens. The percentages of moisture and broken grains were positively correlated (Pearson’s correlation) with the presence of aflatoxin. Milder lesions (grade 1) are positively correlated with aflatoxin and negatively correlated with fumonisin, and the most severe lesions (grade 3) are positively correlated with vomitoxin, fumonisin, and T-2 toxin.

DNA damage by ochratoxin A in rat kidney assessed by the alkaline comet assay BJMBR
Zeljezic,D.; Domijan,A.-M.; Peraica,M..
There are few studies of ochratoxin A (OTA) genotoxicity in experimental animals and the results obtained with cell cultures are inconsistent, although the carcinogenic potential of OTA for the kidney of experimental animals has been well established. We studied the genotoxic potential of OTA in the kidney of adult female Wistar rats (5 in each group) treated intraperitoneally with OTA (0.5 mg kg body weight-1 day-1 for 7, 14, and 21 days) measuring DNA mobility on agarose gel stained with ethidium-bromide using standard alkaline single-cell gel electrophoresis (comet assay). Negative control animals were treated with solvent (Tris buffer, 1.0 mg/kg) and positive control animals were treated with methyl methanesulfonate (40 mg/kg) according to the same...

Effect of fungicide on Fusarium verticillioidesmycelial morphology and fumonisin B1 production BJM
Miguel,Tatiana de Á.; Bordini,Jaqueline G.; Saito,Gervásio H.; Andrade,Célia G.T. de J.; Ono,Mario A.; Hirooka,Elisa Y.; Vizoni,Édio; Ono,Elisabete Y.S..
The effect of fludioxonil + metalaxyl-M on the mycelial morphology, sporulation and fumonisin B1 production by Fusarium verticillioides 103 F was evaluated. Scanning electron microscopy analysis showed that the fungicide caused inhibition of hyphal growth and defects on hyphae morphology such as cell wall disruption, withered hyphae, and excessive septation. In addition, extracellular material around the hyphae was rarely observed in the presence of fludioxonil + metalaxyl-M. While promoting the reduction of mycelial growth, the fungicide increased sporulation of F. verticillioides compared to the control, and the highest production occurred on the 14th day in the treatments and on the 10th day in the control cultures. Fumonisin B1production in the culture...

Effects of fusaric acid on Zea mays L. seedlings Phyton
Diniz,SPSS; Oliveira,RC.
The effects of fusaric acid, a mycotoxin produced by Fusarium sp, were investigated in relation to its action on corn seedlings. The mycotoxin inhibited the development of corn seedlings at 0.2 mM, causing root length reduction. Anatomically, alterations were only visible from 0.5 mM fusaric acid, directly influencing the cell differentiation process. Precocious differentiation reduces the elongation region. This explains (1) that root shortening is morphologically visible, and (2) the differentiation of a great number of lateral roots nearby the apex, which can be seen in longitudinal cuts.

Effects of ochratoxin a on broiler leukocytes Rev. Bras. Ciênc. Avic.
Moura,MA; Machado,CH; Porfírio,LC; Freire,RB.
This study evaluated alterations in the qualitative cellular profile of leukocytes caused by the administration of low doses of ochratoxin-A (OTA) in poultry. Sixty chicks were separated in three experimental groups: control, PBS-treated and OTA-treated. Blood smears from all birds were analyzed three and six hours post-treatment. Differential leukocyte counting demonstrated that OTA reduced the percentage of lymphocytes and eosinophils and significantly increased the number of heterophils and monocytes.

Identification of a splicing coactivator gene that affects the production of ochratoxin a in Aspergillus carbonarius BABT
Lunardi,Lígia Uno; Guembarovski,Roberta Losi; Hanai,Luiz Ricardo; Cristiano,Valderi; Vieira,Maria Lucia Carneiro; Sartori,Daniele; Fungaro,Maria Helena Pelegrinelli.
Ochratoxin A is a mycotoxin produced by some fungi species. Among them, Aspergillus carbonarius is considered a powerful producer. Genes involved in the ochratoxin A biosynthesis pathway have been identified in some producer species. However, there are few studies that purpose to identify these genes in A. carbonarius. The use of insertion mutants to identify genes associated with certain properties has been increased in the literature. In this work, the region of T-DNA integration was investigated in one A. carbonarius ochratoxin-defective mutant previously obtained by Agrobacterium tumefaciens-mediated transformation, in order to find an association between interrupted gene and the biosynthesis of ochratoxin A. The integration occurred in a gene that...

Implementation of a Method to Determine Lolitrem-B in Ryegrass (Lolium perenne L.) by Liquid Chromatography (HPLC) Chilean J. Agric. Res.
Moyano A,Stella; Lanuza,Francisco; Torres B,Alfredo; Cisternas A,Ernesto; Fuentes V,Marcela.
Lolitrem-B is a neurotoxic substance that causes the ryegrass staggers syndrome in cattle, sheep, and horses consuming ryegrass (Lolium perenne L.) infected with endophytic fungus Neotyphodium lolii (Latch, Christensen, and Samuels) Glenn, Bacon, and Hanlin. Due to the increasing use of ryegrass pasture in southern Chile, it is necessary to know the content of lolitrem-B to optimize grazing management and reduce the effect on animal health. The objective of this study was to implement a method to determine lolitrem-B in ryegrass. Ryegrass samples were used without the endophyte, freeze-dried, and kept in darkness at -18 ºC to establish the method. Then, a pure lolitrem-B standard was added and the toxin was extracted with a mixture of chloroform-methanol...

MEIS3 is repressed in A549 lung epithelial cells by deoxynivalenol and the repression contributes to the deleterious effect OAK
Toyotome, Takahito; Takahashi, Hiroki; Kamei, Katsuhiko.
Deoxynivalenol (DON) is an important Fusarium toxin of concern for food safety. The inhalation of powder contaminated with DON is possible and may cause lung toxicity. In this study, we analyzed the gene expression profile of A549 cells treated for 24 hr with 0.2 µg/mL DON by microarray analysis. In total, 16 genes and 5 noncoding RNAs were significantly affected by DON treatment. The repression of B3GALT4, MEIS3, AK7, SEMA3A, KCNMB4, and SCARA5 was confirmed by quantitative PCR. We investigated the DON toxicity on A549 cells that exogenously expressed these 6 genes. The result indicated that A549 cells that transiently expressed MEIS3 were tolerant to the deleterious effects of DON. Our data show that DON affected the expression of genes with various...

Occurrence of aflatoxin B1 in natural products BJM
Prado,Guilherme; Altoé,Aline F.; Gomes,Tatiana C. B.; Leal,Alexandre S.; Morais,Vanessa A. D.; Oliveira,Marize S.; Ferreira,Marli B.; Gomes,Mateus B.; Paschoal,Fabiano N.; Souza,Rafael von S.; Silva,Daniela A.; Madeira,Jovita E. G. Cruz.
The media claims for the consumption of natural resource-based food have gradually increased in both developing and developed countries. The interest in the safety of these products is partially due to the possible presence of toxigenic fungi acting as mycotoxin producers, such as aflatoxins produced during the secondary metabolism of Aspergillus flavus, A. parasiticus and A. nomius. Aflatoxins, mainly aflatoxin B1, are directly associated with liver cancer in human beings. This paper is aimed at evaluating the presence of aflatoxin B1 in a few vegetable drugs, dried plant extracts and industrialized products traded in 2010 in the city of Belo Horizonte, State of Minas Gerais, Brazil. The method used for the quantification of aflatoxin B1 was based on...

Occurrence of killer yeast strains in industrial and clinical yeast isolates Biol. Res.
BAEZA,MARCELO E; SANHUEZA,MARIO A; CIFUENTES,VÍCTOR H.
The secretion of proteinaceous toxins is a widespread characteristic in environmental and laboratory yeast isolates, a phenomenon called "killer system". The killer phenotype (K+) can be encoded by extrachromosomal genetic elements (EGEs) as double stranded DNA or RNA molecules (dsDNA, dsRNA) or in nuclear genes. The spectrum of action and the activity of killer toxins are influenced by temperature, salinity and pH of media. In the present work we determined the existence of K+ in a collection of S. cerevisiae and P. anómala yeasts isolated from environmental, industrial and clinical sources. The assays were performed in strains belonging to three yeast genera used as sensitive cells and under a wide range of pH and temperatures. Approximately 51 % of...
